P450-Glo™ CYP3A4 Assay は発光法を利用してシトクロムP450 CYP3A4 活性を測定するホモジニアスタイプのシステムです。このアッセイはP450 活性への薬剤または新規化合物などの被験物質の影響をテストするために、組換え体またはネイティブソースからの P450酵素の活性を測定するためにデザインされています。この発光アッセイは優れた感度と低いバックグラウンドおよび広いダイナミックレンジを有しています。
ルシフェラーゼ酵素の基質である甲虫ルシフェリンの誘導体である発光 P450 基質を採用しています。この誘導体は P450 により ルシフェリンに変換されることでルシフェラーゼの基質となり、 P450 活性に比例した発光を生じます。
P450の基質であるルシフェリン誘導体と安定化された組換えルシフェラーゼ (Ultra-Glo Luciferase) および特殊なバッファーシステムにより ”グロータイプ (長時間発光)” の発光シグナルを生じます。発光半減期は 2時間以上なのでインジェクターを備えたルミノメーターを使用する必要はなく、プレートのバッチ処理も可能です。また、シトクロム P450 阻害剤スクリーニングでの被験物質のルシフェラーゼ阻害による偽陽性も最小限に抑えられるように最適化されています。
化合物の可溶化に汎用されるジメチルスルホキシド (DMSO) はシトクロム P450 の 3A4 アイソフォーム活性を著しく阻害します（0.1% 以下の低濃度でも）。P450-Glo™ CYP3A4 System (Luciferin-PPXE) DMSO-Tolerant Assay は 3A4 反応において DMSO 耐性を向上させるように特にデザインされています。このアッセイでは DMSOを含まないコントロールと 0.2% DMSO を比べた場合、同等のシグナル / バックグラウンド比が得れます。
