Ribonuclease H (RNase H)は、DNAとハイブリダイズしているRNAのホスホジエステル結合を特異的に加水分解し、3′-末端に水酸基と5′-末端にリン酸基を持つ分解産物を生成するエンドリボヌクレアーゼです。ただし、一本鎖の核酸、二本鎖DNA、二本鎖RNAは消化しません。

Unit Definition: One unit is defined as the amount of enzyme required to produce 1nmol of acid-soluble ribonucleotides from radiolabeled poly(rA):poly(dT) in 20 minutes at 37°C in 20mM HEPES-KOH (pH 7.8), 50mM KCl, 10mM MgCl2, 1mM DTT, 20μM radiolabeled poly(rA):poly(dT).
