T7 RNA Polymerase

T7 RNA Polymeraseは、DNA依存性RNAポリメラーゼで、T7プロモーター配列に極めて高い特異性を示します。

品質検査法：活性試験、SDS-PAGE純度検定、DNase/RNase試験、エンドヌクレアーゼ試験、転写試験

• 特異性： T7 RNA Polymeraseは、T7プロモーターシークエンスに高アフィニティーかつ特異性を示します。
• 高純度： SDS ポリアクリルアミドゲル電気泳動では90%以上の純度を示し、RNase やDNase の活性は認められません。
• 柔軟性： ³H, ³²P, ³³P および³⁵S 標識リボヌクレオシド三リン酸の取込みが可能。
• 5X Reaction Buffer添付：酵素とともに100mM DTTとTranscription Optimized 5×buffer (200mM Tris-HCl (pH7.9 at 25°C), 30mM MgCl₂, 10mM spermidine, 50mM NaCl) が含まれます。

Unit Definition: One unit is defined as the amount of enzyme required to catalyze the incorporation of 5nmol of CTP into acid-insoluble product in 60 minutes at 37°C in a total volume of 100μl. The reaction conditions are: 40mM Tris-HCl (pH 7.9 at 25°C), 6mM MgCl₂,10mM DTT, 10mM NaCl₂, 2mM spermidine, 0.05% Tween® 20, 0.5mM each of ATP, GTP, CTP, and UTP, 0.5μCi [³H]CTP and 2μg supercoiled pGEM®-5Zf(+) Vector DNA.
