リン酸化または総タンパク質の高速定量

Lumit® Immunoassay Cellular System は、細胞溶解液中の標的分子を直接測定可能な洗浄のいらない生物発光イムノアッセイです。このアッセイは、適切な一次抗体（お客様提供）が利用可能であれば、あらゆるタンパク質やリン酸化タンパク質あるいは低分子を検出可能です。

新しい溶解バッファー（Lumit® Lysis Buffer II）を開発し、Lumit® イムノアッセイ細胞システムで解析可能な細胞ターゲットの範囲が拡大されました（下表参照）。

また、pERK, pAKT, pSTAT3, pIkBα, pBTKなどの主要なシグナル伝達ノードを対象として、抗体セットを含むプレデザインキットが early access製品としてご利用いただけます。

Lumit® イムノアッセイ細胞システムには、NanoBiT® サブユニット（LgBiT および SmBiT）と結合したLumit® 二次抗体（ウサギおよびマウス用；ヤギ用のLumit® 抗体コンジュゲートは別売）が含まれています。抗体複合体が結合することで、LgBiT と SmBiT が近接し、活性型の NanoLuc® ルシフェラーゼを形成し、ターゲットアナライトの量に比例した発光を生じます。

ここでは、カーディフ大学医学部のTracey Martin博士が、運動性タンパク質N-WASPの研究においてLumit®イムノアッセイをどのように活用しているかを紹介しています。このプロジェクトでは、N-WASPの発現およびリン酸化状態への影響を評価するために、150種類の新規化合物をスクリーニングしています。それぞれの化合物について複数の濃度を異なる細胞種で試験しており、大量のサンプルが生成されます。当初チームはN-WASP発現量の測定にウェスタンブロッティングを使用していましたが、コストが高く、労力と時間がかかりすぎるという問題がありました。
