迅速で効率的な抗体またはタンパク質の標識
Lumit® Immunoassay Labeling Kit を用いれば、抗体やタンパク質を HaloTag®-SmBiTおよびHaloTag®-LgBiTで標識し、 独自の Lumit®イムノアッセイを開発することができます。Lumit® 検出試薬 A および B は、標識した一次抗体を用いた Lumit® イムノアッセイからの発光シグナルを検出するために特別に設計されています。
Lumit® イムノアッセイは NanoLuc®バイナリーテクノロジー（NanoBiT® テクノロジー）をベースにしています。生体分子相互作用研究用にデザインされた構造相補性レポーターである NanoBiTは、安定性と弱い親和性による最小限の自己会合性について最適化された2つのサブユニット、Large BiT（LgBiT）とSmall BiT（SmBiT）で構成されています。これらのサブユニットを相互作用する2つのタンパク質にそれぞれ融合すると、サブユニットが近接し、発光酵素として再構成されることにより相互作用が起こったことが "報告" されます。
Lumit® Immunoassay Labeling Kitと検出試薬の仕組み
Lumit®イムノアッセイは、HaloTag®テクノロジーを使用したSmBiTおよびLgBiTによる抗体およびタンパク質の化学標識に基づいています。分析物の存在下では、2つの抗体が近接し、SmBiT と LgBiT が活性酵素を形成し、分析物濃度に比例した明るい発光シグナルを生成します。
Lumit® 検出試薬 A および B は、標識された一次抗体を使用する Lumit®イムノアッセイからの発光シグナルを検出するように特別にデザインされています。Lumit® Detection Reagent A は生化学的および細胞ライセートを用いる場合に使用されますが、Lumit® Detection Reagent B は特に、ウシ胎児血清（FBS）を含む細胞培養培地からの検出用、および培地と細胞を含む細胞培養ウェルからの検出用として特別に設計されています。
HaloTag®テクノロジーを使用した化学標識
標識は 2段階のプロセスで行われ、アミン反応性の HaloTag® Succinimidyl Ester（O4）Ligand は、抗体やタンパク質のリジンアミノ酸の第一級アミンと反応します。次に、HaloTag®リガンドで標識された抗体やタンパク質を HaloTag®-LgBiT または HaloTag®-SmBiT 融合タンパク質とインキュベートして、抗体-HaloTag®-LgBiTまたは抗体-HaloTag®-SmBiTの共有結合コンジュゲートを作成します。
シンプルな追加および混合プロトコル、洗浄ステップは不要です！
