Simple, Reliable Plasmid Purification
Wizard® SV 96 and SV 9600 Plasmid DNA Purification Systemはシリカ膜を使った、迅速なプラスミドDNA精製のシンプルで再現性のある方法で、96ウェル、ハイスループットフォーマットです。一枚のプレートの処理時間は60分間かそれ以下です。Vac-Man® 96 Vacuum Manifoldを使うことで、排液の取扱いを無くし、ライセートの清澄化とDNAの結合を同時に行うことができ、サンプル処理中のクロスコンタミネーションを防げます。
精製プラスミドは、そのままDNAシークエンシングや一般的な分子生物学手法に用いることができます。BigDye®ターミネーターによるpGEM®-3Zf(+)ベクターのシークエンシングで98%を超える精度で600塩基以上解析できるという条件を満たす精製プラスミドDNAが得られます。
Wizard® SV 96 and SV 9600 Systemは、手作業あるいは自動分注ロボットを用いて行うことができます。評価済みの自動化方法は、www.promega.com/automethods/ をご参照ください。
このシステムには、真空度50-68kPaの真空ポンプ、Vac-Man® 96 Vacuum Manifold (Cat.# A2291)あるいは同等のマニホールドが必要です。Vac-Man® 96 Vacuum Manifoldを用いる場合、真空トラップを別途ご準備下さい。
