FFPE サンプルからより実用的な RNA を精製

FFPE サンプルより RNA を抽出できれば、病態や組織形態と遺伝子発現を関連付けることができます。しかし、パラフィン固定時にタンパク質と核酸がクロスリンクし、RNA分析を妨げる修飾なども入るため、FFPE 組織からの RNA 抽出は容易ではありません。

ReliaPrep™ FFPE Total RNA Miniprep System はRNA の修飾を戻すために溶解条件が最適化されており、オーバーナイトでの消化も不要です。また、このシステムではキシレンやその他の有機溶媒に頼らない脱パラフィン処理を採用しています。キシレンを用いた脱パラフィン処理にともなう洗浄ステップが無いため、低分子 RNA 断片の保持にも役立っています。

プロトコールは手間も最小限で約1.5 時間で完了します。

この実験では精製した RNA 5µl を鋳型として GoScript™ Reverse Transcription System を用いて逆転写し、cDNA 産物 5µl を用いてマルチプレックス PCRを行った。80bp-290bp の断片が検出できた。
