GoScript™ Reverse Transcription System
このキットには、GoScript™ Reverse Transcriptase, Reaction Buffer, MgCl2, dNTP Mix, Oligo(dT), Random Primers および Recombinant RNasin® Ribonuclease Inhibitorが含まれており、第1鎖cDNAを効率よく合成できるようにデザインされています。トータルRNA、poly(A)+ mRNA または合成転写RNAを始めとするRNAテンプレートを効率よく逆転写します。
GoScript™ Reverse Transcription Mix, Oligo(dT)
GoScript™ Reverse Transcription Mix, Oligo(dT)には、第1鎖cDNAを効率よく合成できるように最適化された試薬セットが含まれています。Oligo(dT)のプライミングは、poly(A)RNA分子の3’末端にアニーリングすることにより開始されます。トータルRNAまたはmRNAの逆転写はどんなpoly(A)+RNAからもoligo(dT)プライム産物となります。この方法を使って生成されるcDNAのポピュレーションは単一の逆転写反応からの数多くの異なるダウンストリームアプリケーションを可能にします。
GoScript™ Reverse Transcription Mix, Random Primers
GoScript™ Reverse Transcription Mix, Random PrimersはqPCR増幅のために準備される第一ストランドcDNA合成のためのランダムプライマーを含んでいます。ランダムプライマーを用いたプライミングは多彩なRNAテンプレートからのcDNA合成を開始する方法として最も一般的なものです。ランダムプライマーはpoly(A)鎖を持たないRNAや原核生物からのRNAを含むすべてのRNA分子から第一ストランドcDNAをプライミングするために使用されます。

逆転写パフォーマンスデータ
以下データはcDNA合成およびRT-qPCRにおけるGoScript Reverse Transferaseの感度、阻害剤耐性、パフォーマンスを示します。
広いRNAインプット量にわたる効率的なcDNA合成
GoTaq 2 step RT-qPCR systemを用いたヒトtotal RNA中の6種の異なるターゲットの定量。log倍に変化させた9点のtotal RNAインプット（10 ng/ul to 0.1 fg/ul）を含む反応液を調製し、GoScript逆転写反応を実施した。各逆転写反応産物10 ul (100 ngから1 fg インプットRNAに相当)を希釈せず直接GoTaq qPCRのサンプルとして使用した。それぞれのターゲット特異的なqPCRアッセイを設計し、かつ以下に示すように相対的な発現レベルが異なるターゲットをアッセイに使用した：extremely high (5.8S rRNA), high (ACTB and B2M), moderate (HPRT ) or low (RPL29 and TBP1 )。Total RNA中におけるそれぞれのターゲット量の違いにともないCq値は異なるが、インプット量のlog変化ごとに≅3.3と理論上に近いサイクル数変化を示す結果が得られている。
阻害剤に対する耐性
In vitro転写で合成したRNAをoligo dTプライマーを用いて逆転写反応させた（反応条件は各製品説明書に準拠）。エタノール非存在下および存在下（5%、10％および20%）で逆転写反応を行い、合成したcDNAをGoTaq qPCR Master Mixを使用してqPCR解析した。エタノール非存在下と存在下のサイクル閾値の変化を転写物レベルの相対変化 ＝ （2Cqinhibitor -Cqnone）に変換した。データは3回の独立した実験から得られた平均値を表している。
低コピーターゲットの効率的な検出
10倍希釈系列に調製した(1µg - 100pg) ヒトtotal RNA (Agilent, Cat #750500)をGoScript™ Reverse Transcription Mix, Random PrimersもしくはOligo(dT)を用いて逆転写反応させた。合成されたcDNAをGoTaq® qPCR Master Mix (Cat. # A6002)を用いて低コピー遺伝子（APCもしくはTBP）に対するqPCR解析を実施した。検量線から求められる増幅効率はMIQEガイドラインで推奨される範囲内であり、それぞれ99% (APC)および97% (TBP)であった。逆転写酵素なし、およびテンプレートなしのコントロールはいずれも増幅ネガティブであった。

GoScript™ Reverse TranscriptaseによるcDNA合成とGoTaq® qPCR Master Mixの組み合わせは、9-logオーダーレンジでテンプレートの検出を可能にする。Panel A. ヒトTotal RNA 10 ng中にMS2 RNAを10倍量ずつ（1 × 102 ～ 1 × 1011コピー）スパイクしたRNAテンプレートを調製した。それぞれのサンプルをGoScript™ Reverse Transcriptaseを用いてcDNA合成し、GoTaq® qPCRで増幅した（n=3）。Panel Aの挿入図は、50, 500, 5,000コピーのスタンダードおよび、5コピーサンプル、テンプレートなしサンプルの解離曲線であり、増幅特異性を示している。Panel Bはそれぞれのサンプルの増幅から求められた検量線を示す。■ = スタンダードサンプル10倍希釈系列; x = 5コピーサンプルの検出結果
