3種類の pBiT3.1 HiBiT Vector は目的タンパク質の N末端または C末端に 11アミノ酸のHiBiT ペプチドタグを付加するために設計された発現ベクターです。これらのベクターにはインフレームの HiBiT 融合タンパク質発現のためのマルチクローニング領域が含まれます。pBiT3.1 Vector は安定あるいは一過性発現の両方に使用することができ、細菌でのセレクション用にカナマイシン耐性遺伝子、哺乳動物セレクション用のブラストサイジン耐性遺伝子をコードしています。目的タンパク質と HiBiT間のリンカーは使用する制限酵素によって長さが変わります。

備考:インサートにはN末端側に ATGコドンまたは Kozak 配列を含む適切な翻訳開始配列が必要で、ストップコドンは含まれてはいけません。
pBiT3.1-secN [CMV/HiBiT/Blast] Vector は膜貫通タンパク質または分泌タンパク質の N末端に HiBiTタグを付加。本ベクターは細胞表面での発現あるいは分泌タンパク質のために HiBiT タグのN末端側へ IL-6 分泌シグナルペプチドをコードしたベクターです。
備考:インサートには翻訳停止のために3´ 末端にストップコドンが必要です。
各種 Nano-Glo® HiBiT detection system と HiBiTペプチドタグを組合わせることで目的タンパク質を発光検出することができます。これにより抗体を必要としないタンパク質の発光検出が可能になります。
備考:
1. HiBiT付きタンパク質の発現は、HiBiTタグと目的遺伝子との間で適切な読み枠が維持されている必要があります。
2. HiBiTペプチド配列は参照用です。HiBiT タグを合成する権利等については使用条件についてご確認ください。

Researcher has no right to modify, derivatize, genetically engineer or otherwise create variations of the NanoBiT® Control Vectors, except that researcher may propagate and store NanoBiT® Control Vectors for use in NanoBiT® assays.
