NanoBRET® スターターシステムのベクターとアッセイデザイン
NanoBRET® PPI MCS Starter System:複数のクローニング部位(MCS)への従来のクローニング手法を用いて、目的のタンパク質にNanoLuc®ルシフェラーゼおよびHaloTag®タンパク質をN末端またはC末端に融合させることができます。
NanoBRET® PPI Flexi® Starter System:目的のタンパク質にNanoLuc®ルシフェラーゼおよびHaloTag®タンパク質をN末端またはC末端に融合させることができます。このクローニングシステムは、2つのレアカッター制限酵素(SgfI および PmeI)を用いた方向性のあるクローニング法で、Flexi® Vector間でタンパク質コード領域を迅速、高精度で効率よく移し換えることができ、シークエンシングによる確認も不要です。NanoLuc® およびHaloTag® 融合タンパク質発現用のFlexi® ORFクローン(Flexi® クローニング対応)ならすぐにPPI実験を開始することができます。

検出装置に求められる機能
NanoBRET® PPI Assay を実施するためには GloMax® Discover のような適切なフィルターを装着でき、2重フィルタリング発光を連続測定できる検出装置が必要です。
ドナーシグナル (例, 蛍光波長 450nm/BP 80nm) を測定するための中心波長が約460nmのバンドパス (BP) フィルターおよびアクセプターシグナル (例, 蛍光波長 610nm/LP) を測定するための約 600–610nm からスタートするロングパス (LP) フィルターが理想的なフィルターセットアップです。この範囲外のフィルターを使用するとデータの質が低下する恐れがあります。
