Enhanced Expression and Solubility of Purified Protein with Novel Tag
HaloTag® Protein Purification Systemは、大腸菌などで発現させる組換えタンパク質の発現量および可溶性を増加させる新規なタグであるHaloTag®の融合タンパク質の精製用にデザ インされています。 HaloTag®テクノロジーでは、目的の タンパク質を効率的、特異的に捕らえ、共有結合により捕捉することができます。HaloTag®タンパク質は 34kDa のヒドロラーゼ変異体で、担体表面に固定されたクロロアルカン-リガンドを介してHaloLink™ Resinに共有結合します。TEV プロテアーゼを用いることでHaloLink™ Resinより標的タンパク質を切り離すことができます。TEV プロテアーゼのN末端にはHQタグ が付加されているのでHisLink™ Resinを用いて容易にプロテアーゼを除去することができます。HaloTag®タンパク質をコードし、大腸菌での発現に適したベクターは、 pFN18A HaloTag® T7 Flexi® Vector（Cat.# G2751）および pFN18K HaloTag® T7 Flexi® Vector（Cat.# G2681）です（キットには付属しません）。
HaloLink®テクノロジーは迅速、便利にHaloTag®融合タンパク質の発現を確認でき、同様にHaloTag® TMR Ligandで蛍光標識した後SDS-PAGEを行うことでHaloLink™ Resinへの固定化効率もモニターできます。ガイドライン： HaloLink™ Resin Technical Manual #TM250、HaloLink™ Protein Array Technical Manual #TM310、HaloCHIP™ System Technical Manual #TM075.
- Hisタグ、GSTタグ、MBPタグよりも優れたタンパク質収量、純度、活性：HaloTag®融合タンパク質がHaloLink™ Resinにより特異的に捕らえられ共有結合
