パスウェイレポーターの構築には、それぞれ異なる組換えホタルルシフェラーゼ遺伝子 (luc2, luc2P, luc2CP) を持つ3種類の 最小限プロモーター(minP) ベクターがご利用いただけます。Luc2遺伝子からはほとんどの変則的に働く転写因子結合サイトが除去され、コドンの最適化により哺乳動物での発現が向上しました。luc2P、luc2CP およびRapidResponse™ 遺伝子はluc2遺伝子にルシフェラーゼの細胞内半減期に影響を及ぼす分解配列を付加したものです。RapidResponse™ 遺伝子は刺激に対してより素早く応答しますが、シグナル強度を犠牲にしています。luc2P遺伝子(半減期 1時間)はluc2(半減期 3時間)よりもより早く応答しますが、中程度のシグナル強度を示し、luc2CP(半減期 0.4時間)はさらに迅速に応答しますが、シグナル強度は低く抑えられています。このminPベクターは選択マーカー(ハイグロマイシン)有/無の2タイプがご利用いただけます。研究をスピードアップするために、すでに応答配列を組み込んだpGL4.27ベースのプレデザインされたパスウェイベクターをいくつか揃えています。これらのいくつかは安定細胞株 (GloResponse™ Cell Lines) もご用意しています。

About the pGL4 Reporter Vectors
The pGL4 Vectors offer increased reporter gene expression with codon optimization of synthetic genes for mammalian expression and reduced background and risk of expression artifacts with removal of cryptic DNA regulatory elements and transcription factor binding sites. Firefly luciferase has options that offer improved temporal response with the destabilized Rapid Response™ luciferase genes.
