RNasin® Plus RNase Inhibitor は反応液の調整中にリボヌクレアーゼを阻害します。反応液を70℃、15分間加熱してもリボヌクレアーゼ活性が再び現れることはありません。したがって、RNasin® PlusをRT-PCRの前にあらかじめ添加することで逆転写反応中のRNAの保護を強化することができます。

RNasin® Plusは高温でも安定
RNasin® Plus RNase Inhibitorは、RNase AやRNase Bのような真核生物のRNaseを阻害することができる組換え哺乳類RNase阻害剤であり、このタンパク質のヒトバージョンと類似している。ヒト胎盤RNase阻害剤に存在する特定のシステイン残基は、その酵素の酸化感受性に関与している(1)。RNasin® Plus RNase Inhibitorはこれらのシステイン残基を含まないため、酸化的損傷に対してより耐性がある。RNasin® Plus RNase InhibitorはRNaseと安定な複合体を形成し、RNaseを不活性化する。この複合体は70℃まで少なくとも15分間安定である (Figure 1)。
RNasin® PlusはRT-PCRで試験されており、ヒトタンパク質と同様に、AMV、M-MLV、ImProm-II™逆転写酵素、TaqおよびTfl DNAポリメラーゼなどの酵素に適合します。また、RNasin® Plusは、TaqMan®アッセイにおける定量的リアルタイムRT-PCRにも適合することが試験で確認されており、ラット肝タンパク質抽出液のような複雑な生物学的抽出液においてもテンプレートを分解から保護することができ、幅広いRNase阻害スペクトルを示している(2)。

Unit Definition
One unit is defined as the amount of RNasin® Ribonuclease Inhibitor required to inhibit the activity of 5ng of ribonuclease A by 50%. Activity is measured by the inhibition of hydrolysis of cytidine 2,3´-cyclic monophosphate by ribonuclease A.
Storage Buffer
20mM HEPES-KOH (pH 7.6 at 4°C), 50mM KCl, 8mM DTT and 50% (v/v) glycerol.
