Lumit™ IL-1βImmunoassayは、シンプルな無洗浄プロトコルを使用して、細胞培養サンプル中のIL-1βを定量的に測定します。サンプルに標識抗体、検出試薬を加え、標準のプレートフォーマットルミノメーターを使用して発光シグナルを測定するだけです。プロトコル全体が70分以内に完了します！このアッセイは、培養中の細胞でそのまま、あるいは培地を別のアッセイプレートに移して測定することができます。
Lumit™IL-1βイムノアッセイのしくみ
IL-1βに対する一次抗体は、特異的かつ高感度な検出を行うために選別され、NanoBiT®ルシフェラーゼのLgBiTおよびSmBiTサブユニットで標識されています。IL-1βの存在下で、サブユニットが会合し、活性を有するルシフェラーゼ酵素を形成します。最適化された基質を添加すると、IL-1βレベルに比例した明るい発光シグナルが生成されます。

Lumit™ IL-1βイムノアッセイの試薬添加＆測定だけのシンプルなフォーマットにより、IL-1β放出モジュレーターのハイスループットスクリーニングが可能。
NLRP3インフラマソーム阻害剤であるMCC950は、カスパーゼ1の活性化と成熟IL-1βの放出を阻害。分化したTHP-1細胞（5×10 4 /ウェル）にMCC950の段階希釈液を加え、続いてLPS（20 EU / ml）またはビークルを加えました。Lumit™IL-1β（ヒト）イムノアッセイは5時間後に実施。
