HaloTag® リガンドによる最適化されたタンパク質精製と捕捉
プロメガの HaloTag® リガンド製品群は、タンパク質精製におけるアフィニティ標識をはじめとした多様な機能に対応しています。これらのリガンドは、生理的条件下で HaloTag® 融合タンパク質と極めて特異的かつほぼ不可逆的な共有結合を迅速に形成し、安定かつ効率的な標識を可能にします。さらに、バックグラウンドの干渉が最小限に抑えられる点も特長です。HaloTag® リガンドを用いることで、適切な化学基を持つ任意の化合物や表面を対象とした解析が可能になります。また、多彩なHaloTag® Ligand Building Blocksが取り揃えられており、応用の可能性は無限に広がります。
HaloTag® Biotin Ligandは、タンパク質の捕捉および解析において、効率的なビオチン標識を可能にします。このリガンドは細胞膜透過性があるため、細胞内タンパク質に対する標識も可能です。インキュベーション後には、未結合のリガンドを洗い流すことができるため、バックグラウンドを抑えた検出が行えます。
プルダウン性能をさらに高めたい場合には、HaloTag® PEG-Biotin Ligandがおすすめです。このリガンドは、柔軟性の高い長めのスペーサー構造を有しており、ストレプトアビジンとの結合効率が優れています。細胞膜透過性が限られるため、標識には細胞溶解処理が必要ですが、この延長スペーサーによって、固定化や誘導体化の過程において HaloTag® 融合タンパク質の活性が保持されやすいという利点があります

HaloTag® 融合タンパク質のゲル解析
p65-HaloTag® 融合タンパク質の効率的な捕捉と、ストレプトアビジンコート磁性粒子での IκB の共捕捉 p65-HaloTag® 融合タンパク質をコードするプラスミドを一過性発現させた CHO-K1 細胞を、HaloTag® Biotin Ligand（25µM）の存在下または非存在下でインキュベートした。細胞溶解後、ビオチン標識されたタンパク質を ストレプトアビジンコート磁性粒子であるMagneSphere® Paramagnetic Particles（カタログ番号 Z5481）で捕捉した。タンパク質は SDS-PAGE にて分離後、ウエスタンブロットにより検出した。一次抗体には p65 抗体（BD Biosciences、パネルA）または IκB 抗体（BD Biosciences、パネルB）を使用した。パネルAおよびBにおける各レーンの内容は以下の通り：C = 細胞溶解液、UB = 非結合タンパク質、B = 結合タンパク質。
