3種類の HiBiT CMV-neo Flexi® Vector は標的タンパク質の N末端または C末端に HiBiT ペプチドタグを付加するために、Flexi® Cloning System (Cat.# C8640) を用いて簡便で効率よく目的遺伝子をクローニングするために設計された発現ベクターです。ベクターは安定または一過性の遺伝子発現に使用でき、バクテリアでのセレクション用のカナマイシン、哺乳動物でのセレクション用のネオマイシンをコードしています。

本ベクターは細胞表面での発現あるいは分泌タンパク質のために HiBiT タグのN末端側へ IL-6 分泌シグナルペプチドをコードしたベクターです。
備考: HiBiTタグタンパク質を効率的に細胞表面で発現させるために、目的遺伝子に本来コードされた分泌シグナルを除去することを推奨しています。
備考:
1. HiBiT付きタンパク質の発現は、HiBiTタグと目的遺伝子との間で適切な読み枠が維持されている必要があります。
2. HiBiTペプチド配列は参照用です。HiBiT タグを合成する権利等については使用条件についてご確認ください。

Flexi® Vector System について
Flexi® Vector System はタンパク質コード配列のディレクショナルクローニング (directional cloning) を行うシンプルで強力なシステムです。2種類のレアカッター制限酵素 SgfI および PmeI による消化をベースにしており、様々な Flexi® Vector 間でのタンパク質コード領域を迅速、効率的、高い忠実性で移換えられるので、シークエンスの確認も不要です。組換えクローンをそのまま使用できるため、バックグラウンドコロニーよりスクリーニングする時間を節約することができます。 また、Flexi® system の汎用性により様々な発現系や融合タグより選ぶことができます。ダイレクトな組換えは2つのN末端タグベクター間またはN末端ベクターからC末端ベクターで行うことができます。
備考:サブクローニングを行う上で、遺伝子が挿入されていないバックグラウンドコロニーを低減するためにFlexi® Vector にはバーナーゼ致死遺伝子が含まれています。Flexi® Vector Cloning System ではバーナーゼ遺伝子と目的遺伝子が置き換わります。購入した空の Flexi® ベクターは標準的な実験で用いられる大腸菌株では増やすことはできません。
