FuGENE® 4K reagent delivers improved DNA transfection performance while minimizing impact on cell health — **FuGENE® 4K試薬は、細胞の健康への影響を最小限に抑えながら、DNAトランスフェクションのパフォーマンスを向上させます。**A375、CHO-K1、H1299、HCT116細胞株で、FuGENE® 4KまたはLipofectamine™ 3000（ThermoFisher Scientific）を用いて試薬：DNA比を変えながらGFP発現コンストラクトをトランスフェクションした。48時間後、底面が透明なプレート内の細胞の総GFP蛍光を測定した。細胞生存率は、CellTiter-Glo® Luminescent Cell Viability Assayを使用して、2枚のプレートで測定した。

FuGENE® 4K vs Lipofectamine™ 3000 Cell Viability — **FuGENE® 4K 試薬は、多くの細胞株で Lipofectamine™ 3000 よりも優れた性能を示しました。**様々な細胞株を播種し、FuGENE® 4KまたはLipofectamine™ 3000トランスフェクション試薬を用いてGFP発現プラスミドをトランスフェクションしました。トランスフェクションの効率は、フローサイトメトリーによりGFPを発現する細胞をカウントして決定した。細胞生存率は、propidium iodide dyeを用いて決定した。

N2A細胞（マウス）に使える新しいプラスミドトランスフェクション試薬を探しており、FuGENE 4Kを試したところ初回の導入で素晴らしい結果が得られ、操作もシンプルでした。ワンチューブで完結するシングル試薬構成である点も魅力で、他社の選択肢と比較しても、まずはFuGENE® 4Kを試すことに迷いはありませんでした。今後、当研究室ではこの試薬を定番トランスフェクション試薬に決定です。
